Brief description:

This new Nipkow spinning disk setup is equipped with the latest Yokogawa Confocal Scanner Unit CSU-W1-T2 SoRa. It contains two interchangeable spinning disks

  1. Disk 1 format: 50 µm pinholes @ 500 µm spacing, dual disk technology for normal spinning disk imaging
  2. Disk 2 format: 50 µm pinholes (SoRa) @ 500 µm spacing, dual disk technolog for "super resolution imaging" XY resolution ~120–150 nm)

Due to the new disk design it almost reachs the z-resolution of a point scanning confocal. It is equipped with 2x back illuminated sCMOS Hamamtasu Orca Fusion BT cameras (2304 x 2304 pixel, 6.5 x 6.5 μm pixel size) that allows big field-of-views and fast and sensitive live cell imaging even in a dual camera mode.

In addition, it is also equipped with a point scanner for FRAP for doing precise FRAP and ablation experiments. It has an full box incubator for temperature and CO2, a Piezo z-​stage (Mad City Labs Nano-​Drive, 200 µm, with triggering) and filter combinations for fast multicolor acquisition. It has also a hardware autofocus system (Nikon Perfect Focus System). The two sCMOS cameras allows for simultaneous acquisition of combinations of blue and yellow, green and red, and blue and far red fluorophores.

The system can also be used for simple widefield imaging.

Furthermore, the system is equipped with a point scanning  "confocal arm", using the new technology of external pageRe-​scan with the external pageRCM system from confocal.nl.

Software:

  • Nikon NIS Elements: The microscope can run all common types of live-​cell experiment, combining time lapse, multi-​position (XYZ), multi-​channel and stitching (large image) functionalities. In addition, it can be custom-​scripted to perform more complex and/or ‘smart’ imaging routines (JOBS). Combined with the larger field-​of-view (FOV) spinning disk unit and triggered piezo Z-​stage, the system is particularly suitable for faster and/or higher throughput confocal imaging.
